Antioxidant and Anti-Inflammatory Activities of Phenolic Acid-Rich Extract from Hairy Roots of Dracocephalum moldavica

This study evaluates the antioxidant properties and anti-inflammatory potential of polyphenolic acid-rich fractions 80% methanolic extract from hairy roots Dracocephalum moldavica. The fractionation crude yielded following: a diethyl ether fraction rich in caffeic acid (DM1) (25.85 mg/g DWE), an n-butyl rosmarinic (DM3) (43.94 DWE) water residue salvianolic B (DM4) (51.46 DWE). content these compounds was determined using high-performance liquid chromatography (HPLC). Their activity evaluated based on DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt) FRAP assays. by their effect nuclear factor kappa-B (NF-κB) activation interleukin-1β (IL-1β) production LPS E. coli stimulated monocytes. level pro-inflammatory IL-1β cells measured ELISA. NF-κB THP1-Blue™ cells, resulting secretion SEAP (secreted embryonic alkaline phosphatase), detected spectrophotometrically Quanti-Blue reagent. Among tested fractions, showed highest potential, with EC50 value 15.41 µg/mL assay 11.47 reduction 10.9 mM Fe(II)/g DWE FRAP. DM1 at concentration 10 mg/mL also efficiently reduced LPS-induced (53% inhibition) (47% without affecting normal growth L929 fibroblast cells.
